How are plays measured? On click or after x seconds?

They are measured on unique clicks.

I know there was something in the "like" module to prevent people from boosting likes. Is there anything similar in in the play count?

Yeah there is - it uses several things under the hood (ip tracking, user account, agent and cookie) to try to prevent it from being gamed. Of course nothing is 100% fool proof, but it should work good.
